Although I killed required monster 2 times I cannot finish the quest.

Hey wildfreak, you need to kill a specific one as indicated by the NPC, not just any one that you get from a normal spawn. Move around the front-right part of the boat and you'll find it.

The quest reward says I gain XP in leadership. There is no such skill.

You can not train leadership manually with turns, but it is a general skill in your character sheet.
